减毒沙门氏菌VNP20009联合ADT-OH抗肿瘤效果及其相关机制的研究文献综述

拟研究的问题基因改造过的沙门氏菌VNP20009具有低毒性和高肿瘤靶向性,可显著抑制肿瘤的生长,与此同时硫化氢亦与肿瘤的发生发展密切相关。

本课题拟研究通过使用有机硫化氢供体ADT-OH与VNP20009联用来评价其对B16F10小鼠黑色素瘤的生长抑制效果并解释其抑制的可能相关机制,为治疗黑色素瘤提供研究方向。

研究手段1.F10小鼠黑色素瘤模型构建F10细胞用DMEM培养基培养至指数生长期,用0.5%胰酶消化,800rpm/min 离心5min,PBS 洗涤2次后,细胞计数,用PBS 重悬细胞,将F10调整为1106个/mL。

每只小鼠接种100mu;L于C57BL/6小鼠腋下脂肪垫处,即F10接种1105个/只。

接种小鼠饲养于清洁级动物房,待肿瘤生长至体积约150 mm3 时进行后续实验。

2.VNP联合ADT-OH对F10B16小鼠黑色素瘤生长抑制效果分析如前所述,接种F10的荷瘤小鼠肿瘤体积生长至150 mm3左右时,随机分为PBS 组、VNP 组、ADT-OH组、VNP ADT-OH组,测定每组小鼠肿瘤体积,作为给药前肿瘤体积。

随后PBS 组、VNP 组、VNP ADT-OH组分别腹腔注射100ul PBS、VNP沙门氏菌,菌体量1105 cfu,之后每隔一天测定肿瘤体积,详细记录小鼠在治疗过程中存活情况。
