1. 研究目的与意义(文献综述)
透明质酸(hyaluronic acid,ha)是糖胺聚糖中的一种,属于酸性粘多糖,广泛分布于人体各部位,其中皮肤也含有大量的透明质酸。
1934年美国哥伦比亚大学教授meyer等人首先从牛眼玻璃体中分离出透明质酸。
在机体内,透明质酸是一种多功能基质,显示出多种重要的生理功能,例如调节蛋白质,协助水电解质的扩散及运转,润滑关节,调节血管壁的通透性,促进伤口愈合等等。
2. 研究的基本内容与方案
主要内容: 1.ha的脱乙酰化最佳工艺条件筛选; 2.相关产物的表征和性能测试技术方案: 乙酰氨基交联:ha除了有活性的羧基和羟基外,还有可进行改性的乙酰氨基,亦可进行交联。
ha脱乙酰基作用后有游离氨基,vittorio crescenzi等对ha脱酰基化及硫酸盐化进行了研究。
ha部分脱酰基化(deha):55℃下,在一定量的ha中加入水合肼及其硫酸盐搅拌72h,加入低温乙醇沉淀,冲洗,真空干燥24h得到聚合物产品。
3. 研究计划与安排
第1-2周:查阅相关文献资料,明确研究内容,拟定试验方法和条件。确定方案,完成开题报告。
第3-12周:完成ha的脱乙酰化。
第13-14周:完成并修改毕业论文。
